Hi folks,
For RDC measurement with polyacrylamide gels, by using the conventional
IPAP [15N, 1H]-HSQC pulse scheme proposed in 1998 by Ad Bax and coll.
{JMR v131, 373-378 (1998)}, when protein concentration is too low, NH2
groups from polyacrylamide gel can interfere with the NH groups from
protein.
Robet Tycko and coll. described 3 years later a modified IPAP HSQC pulse
scheme designed to suppress natural-abundance 13N signals from NH2
groups of polyacrylamide gels {/Journal of Biomolecular NMR v21, 141-151
(2001)}/.
I am very intersted by this modified version but this pulse sequence is
not find in the Agilent vnmrj library.
So before to start to spend time to rewrite/edit the gNhsqc_IPAPA.c I
would like to know :
a) your feedback on this pulse sequence (do you really seen a dramatic
decrease of the gel signals and without decrease of the S/N for the
protein signals)
and
b) if somebody have already this pulse sequence in the vnmrs format it
will be very cool to share it !!!
