We have recently acquired a Bruker 500 MHz instrument with a QNP cryoprobe and when we run 2D noesy we do not see any meaningful crosspeaks.
If we run a 2D noesy in our old 500 MHz instrument we get a perfect spectrum with all crosspeaks. If we run the same experiment and sample in the new 500 MHz instrument with cryoprobe we see no crosspeaks at all. 2D roesy is not affected and works OK in both instruments. The 1D experiments show the same problem (no signals for noesy, no problem with roesy).
We have tried every possible parameter, pulse program, etc. available. The only difference is that the old instrument runs Topspin 2.1 and the new one runs Topspin 3.0.
Has anybody experienced similar problems with noesy under Topspin 3.0? We are pretty at a loss, any comment or information will be welcome.
