Hello AMMRL group,
I am trying to acquire 15N-gHSQC using Broadband probe. I have ubiquitin sample (90%H2O/10%D2O). I am not sure if I should change the filter for 15N for BB probe? I also have HCN triax probe, but one faculty prefer to use Broadband to get 15N-HSQC. So, I am not sure what the setup is. I usually change the quarter loop and filter for triax. We also have Biopack but this person prefer the BB with regular 15N-HSQC, so I am trying to help him out. Has anyone acquire 15N using broadband probe?
Here are the steps I took to set up 15N-gHSQC using BB probe.
1) Change only the quarter loop, keep the broband filter.
2) X-tune to 15N frequency.
3) Tune proton.
4) Set up experiment for N15-HSQC. Change the F1 Nucleus to 15N. Didn't get any signal.
Any suggestions will be helpful. Thank you.
