AMMRL: Help with 15N NMR

Hi Folks,

I hope everyone is safe. I miss the days when running out of helium was our biggest concern.

My institution has no on-site NMR expert, and as such I find myself in need of expertise. I am working with a Varian 500 running VNMR software. We have fully disabled autotuning due to two very expensive probe repairs related to failure of the end-stop. I believe the instrument is currently configured such that channel 1 is tuned to 1H, and channel 2 is tuned to 15N instead of 13C. I am interested in collecting 15N spectra on a sample containing 25 mM of commercial 15N-labeled phenylalanine dissolved in D2O. I have collected a high-quality 1H spectrum, but my initial attempt to collect a 15N spectrum with standard Varian parameters shows only noise going all the way out to 1000 ppm. I suspect this is because I have no idea what I’m doing. If anyone with expertise in Varian hardware and software would be willing to help me troubleshoot this, ideally while I’m sitting at the console on a Monday, Wednesday or Friday, I would be eternally grateful.
