We are in the process of upgrading our AMX-500 to z-axis PFG capabilities
and had planned to just upgrade our existing Bruker 5mm dual and 5mm inverse
probes (circa 1991) by having Bruker add the z-gradient coils to them. We
have however been informed that these probes are too old to upgrade. We are
now in the dilemma of whether to buy a new 5mm dual gradient probe or just
buy a 2.5 mm (or 3mm:>) dual gradient probe since we planned to buy one
eventually to increase our 13C detection sensitivity. We are a natural
products group and typically have from 0.5 mg to 10 mg of small molecule (MW
<1000) to get our structures. Solubility is usually not a problem. My
question is: what are the pros and cons of using a dual microprobe as the
routine probe as opposed to a 5mm probe. In particular, is it more
difficult to shim? Are the NMR tubes more expensive? What other things do
I need to take into consideration? Thanks for any insight on this. I will
post a summary of responses.
