We have a Inova-500 and VNMR61C. We have been working on cell wall structures with Varian's nanoprobe with whole cells, mainly use HSQC. One thing we want to make sure is that the signals that we see on the HSQC are really from the cell walls which has relatively slow diffusion, rather than from metabolites which have much faster diffusion rate. I know Dr. Shapiro's group in Lilly has developed some pulse sequence in Bruker's machine to suppress the very broad peak by adding a CPMG to the HSQC sequence, while at the same time, use gradient to remove signals from small, fast moving molecules. I am just wondering if anyone has a Varian version pulse sequence to do that. Idealy it would be a modified HSQC or gHSQC sequence which contains a CPMG filter for very broad peaks as well as a gradient filter to avoid the fast diffusing metabolites, but we will be happy even if it only has the gradient part to filter out the small metabolite.
